Overview

The MicroRNA 10b precursor (pre-miR-10b) is a non-coding RNA hairpin structure that serves as the immediate precursor to mature miR-10b, a well-characterized oncomiR associated with high metastatic potential in various cancers. In malignancies such as metastatic breast cancer and glioblastoma, miR-10b is frequently overexpressed and functions by suppressing the translation of Homeobox D10 (HOXD10), which in turn leads to the upregulation of the pro-metastatic gene RHOC (Ma et al., 2007, Nature). The precursor hairpin is a significant therapeutic target because its unique secondary structure allows for the design of small molecules and antisense oligonucleotides that can specifically inhibit its maturation. For instance, the small molecule Targapremir-10b binds to the Dicer processing site of the hairpin, preventing the production of mature miR-10b and effectively reducing tumor cell invasion (Costales et al., 2020, Science Advances). Beyond its role in oncology, dysregulation of miR-10b has also been linked to neurodegenerative diseases like Huntington's disease, suggesting a broader clinical relevance (Hoss et al., 2014, Frontiers in Aging Neuroscience). Consequently, targeting the pre-miR-10b hairpin represents a precision medicine approach to modulating gene expression in complex disease states.
