Overview

MicroRNA 1229 (MIR1229) is a non-coding RNA molecule that regulates gene expression post-transcriptionally by binding to target mRNA transcripts, impacting their stability and translation[3]. It is processed from a primary transcript (pri-miRNA) to a precursor hairpin (pre-miRNA) and finally to a mature miRNA form, which is loaded onto the RNA-induced silencing complex (RISC) and guides it to specific mRNAs for silencing[3]. MIR1229 has been associated with diseases such as colorectal adenoma and is implicated in regulatory processes in cancers such as breast and gastrointestinal cancer (where it may contribute to chemoresistance)[2][3]. In neurodegenerative contexts, specifically Alzheimer’s disease, a variant of pre-miRNA-1229 (rs2291418) is linked to altered structure formation and increased mature miR-1229-3p levels, which regulate the translation of the SORL1 gene involved in amyloid precursor protein (APP) sorting, suggesting its potential as a therapeutic target[1]. MIR1229 is also being studied as a circulating biomarker for prognostic purposes, particularly for chemoresistance in cancer[2].
