MicroRNA-1258 (miR-1258) is a small, non-coding RNA approximately 21–22 nucleotides in length, located within the first intron of the ZNF385B gene on chromosome 2q31.3[1][2]. It functions mainly as a post-transcriptional regulator by binding complementary sequences in the 3' untranslated regions of specific mRNAs, leading to their degradation or translational repression[1][2]. miR-1258 is frequently downregulated in multiple cancers, where it acts as a tumor suppressor by inhibiting cell proliferation, migration, invasion, cell stemness, glycolysis, and facilitating apoptosis[1][2][3][4]. Its tumor-suppressive actions are mediated through direct targeting of genes such as RPN2, E2F1, CKS1B, GRB2, among many others, and by modulating key signaling pathways like the TGF-β/Smad and GRB2/Ras/Erk pathways[1][2][3][4]. miR-1258 is being investigated as a biomarker for cancer diagnosis, prognosis, and potentially as a therapeutic target, but no drugs are currently approved that directly target this miRNA[1][2][4].
