Overview

MicroRNA 135b (miR-135b) is a short, non-coding RNA molecule of the microRNA family, transcribed from the MIR135B gene on chromosome 1q32.1. It functions as a regulator of gene expression by binding to mRNAs and inhibiting their translation or promoting degradation. miR-135b is upregulated in a variety of cancers, including colorectal, breast (especially triple-negative subtype), lung, liver, and others, where it promotes oncogenic phenotypes such as increased cell proliferation, migration, invasion, angiogenesis, and drug resistance. It interacts with several key pathways, including Hippo, Wnt, TGF-β, and ERBB signaling. Dysregulation of miR-135b is also implicated in non-cancer diseases such as cardiovascular and neurodegenerative conditions. Its expression is controlled by transcription factors, signaling pathways, epigenetic modifications, and non-coding RNAs. As both a disease biomarker and a potential therapeutic target, miR-135b is of intense scientific and clinical interest. Manipulation of its expression or activity holds promise but may present safety challenges because of its broad impact on gene regulation.

Mechanism of action

Antagomirs and sponges inhibit miR-135b, which can restore tumor suppressor pathways and reduce migration, invasion, and metastasis in cancer models. Certain drugs and natural compounds regulate upstream expression of miR-135b.
