Overview

MicroRNA-154 is a small non-coding RNA molecule predominantly involved in the post-transcriptional regulation of gene expression via mRNA silencing. It functions largely as a tumor suppressor in various cancers, including bladder, breast, colorectal, prostate, and liver cancers, where its expression is frequently downregulated. miR-154 directly targets mRNAs such as ATG7, E2F5, ZEB2, RSF1, and RUNX2, inhibiting cell proliferation, migration, invasion, and tumor growth. Its reduced expression correlates with poor clinical outcomes and more aggressive disease phenotypes, making it a potential therapeutic target and prognostic biomarker. Research is ongoing into miRNA-based therapies to restore its function, although challenges remain in drug delivery and specificity
