Overview

MicroRNA 181a (miR-181a) is a small non-coding RNA molecule that functions as a key post-transcriptional regulator of gene expression by binding to the 3' untranslated regions of target messenger RNAs. It is highly expressed in the thymus and plays a critical role in the immune system, where it modulates T-cell receptor sensitivity and signal transduction by targeting various phosphatases such as DUSP6 and SHP-2 (Li et al., 2007, Nature). Beyond its immunological functions, miR-181a is involved in regulating cell proliferation, apoptosis, and differentiation across multiple tissue types, often by targeting the anti-apoptotic protein BCL-2 (Ouyang et al., 2012, Journal of Cerebral Blood Flow & Metabolism). In the context of disease, miR-181a exhibits a dual role in oncology, acting as an oncomiR in certain leukemias while functioning as a tumor suppressor in solid tumors like gliomas (Shi et al., 2008, Gene Therapy). It is also implicated in cardiovascular diseases, where it regulates vascular inflammation and atherosclerosis (Indrieri et al., 2020, International Journal of Molecular Sciences). Therapeutic strategies currently under investigation include the use of synthetic miR-181a mimics to restore its function in depleted states or antagomirs to inhibit its activity when it is pathologically overexpressed. Although primarily in the preclinical research stage, miR-181a represents a promising target for the development of precision medicines for hematological malignancies and inflammatory disorders.
