MicroRNA 181a-2 (MIR181A2) is a non-coding RNA classified as a microRNA, functioning as a post-transcriptional regulator of gene expression. MIR181A2 is part of the miR-181 family, regulating cell fate, differentiation, apoptosis, mitochondrial function, and immune homeostasis. It acts as an oncogene in several cancers, especially gastric and hepatic carcinomas, through repression of target mRNAs such as CAPRIN1 and MYLK, thereby promoting proliferation, survival, and metastasis. Its altered expression is also implicated in neurodegenerative disorders and embryonic development. MIR181A2 is under investigation as a therapeutic target and biomarker; however, due to its pleiotropic effects, therapeutic development faces significant specificity and safety challenges.
Therapeutic strategies are experimental and include antisense oligonucleotides or inhibitors to suppress miR-181a-2 function, aiming to restore tumor suppressor activity or reverse chemoresistance. Drug resistance mechanisms may be modulated via target gene interactions (e.g., TIMP3, MYLK).
