Overview

MicroRNA 181b-1 (miR-181b-1) is a member of the miR-181 family, classified as a small, single-stranded noncoding RNA molecule that regulates gene expression primarily via post-transcriptional mechanisms. It exerts important biological effects in the vascular endothelium by targeting mRNAs such as importin-α3, which is required for NF-κB nuclear translocation. By doing so, miR-181b-1 inhibits NF-κB–mediated inflammatory signaling, reduces expression of adhesion molecules (VCAM-1, E-selectin, ICAM-1), and limits leukocyte adhesion and transmigration. Clinically, dysregulation of miR-181b-1 is implicated in vascular inflammation, sepsis, cardiovascular diseases, and several cancers, especially breast cancer where high expression is associated with aggressive, poor-prognosis subtypes. Therapeutic manipulation (mimics or inhibitors) of miR-181b-1 has shown promise in preclinical models for modulating inflammation and tumor progression, but no approved drugs are currently available. Its potential as both a biomarker and therapeutic target is the subject of active investigation[1][2].
