Overview

MicroRNA 194-3p (miR-194-3p) is a mature, small non-coding RNA molecule that functions as a critical post-transcriptional regulator of gene expression. It is typically processed from the miR-194-1 or miR-194-2 precursors and is often co-expressed with miR-192 or miR-215 in a tissue-specific manner, particularly within the gastrointestinal tract and liver (miRBase, 2023). miR-194-3p exerts its biological effects by targeting the 3' untranslated regions of specific mRNAs, such as those encoding for oncogenes or tumor suppressors, thereby influencing pathways like Wnt/beta-catenin and PI3K/Akt (PubMed, 2021). In clinical contexts, miR-194-3p has been identified as a potential biomarker and therapeutic target in various malignancies, including gastric and colorectal cancers, where its dysregulation correlates with tumor progression and metastasis (NCBI, 2022). It also plays roles in non-oncogenic conditions such as osteoarthritis and renal fibrosis by modulating inflammatory and fibrotic signaling (PubMed, 2019). While no clinical drugs are currently approved, experimental approaches using miRNA mimics or antisense inhibitors are being explored to modulate its activity in disease states (Journal of Hematology & Oncology, 2020). The therapeutic potential of miR-194-3p lies in its ability to simultaneously regulate multiple genes within a single signaling network, though delivery to target tissues remains a significant challenge.
