MicroRNA 196b (miR-196b) is a member of the miR-196 gene family, encoded within the HOXA gene cluster on chromosome 7p15.2 in humans. It acts by suppressing target messenger RNAs through the RNA-induced silencing complex (RISC), leading to translational inhibition or mRNA degradation. miR-196b tightly regulates genes critical for embryonic development, hematopoiesis, and apoptosis. In cancer, dysregulation of miR-196b promotes abnormal proliferation and survival, contributing to malignancy, particularly in MLL-rearranged leukemia, where its overexpression leads to a block in hematopoietic differentiation and enhanced proliferation. Conversely, in some contexts, miR-196b may act as a tumor suppressor by repressing the expression of oncogenic HOX genes. Its levels in patient tissue samples can serve as a prognostic biomarker, and modifications targeting its activity (e.g., antagomirs) are being explored preclinically as therapeutic approaches.
Antagomir or antisense inhibition of miR-196b blocks its activity, affecting cell proliferation and differentiation in leukemia. Potential for miRNA mimic or replacement therapy in contexts where miR-196b acts as a tumor suppressor.
