Overview

MicroRNA 196b-5p is a small non-coding RNA molecule that functions as a critical post-transcriptional regulator of gene expression. It is encoded within the HOX gene clusters, specifically between HOXA9 and HOXA10, and is frequently co-expressed with these developmental regulators (PubMed: 17339450). In the context of oncology, miR-196b-5p often functions as an oncomiR, particularly in MLL-rearranged acute myeloid leukemia and glioblastoma, where its overexpression promotes cell survival and inhibits apoptosis by targeting genes like FAS and IκBα (PubMed: 21602825, PubMed: 24553118). However, its role can be context-dependent, occasionally acting as a tumor suppressor in specific solid tumors. Therapeutic interventions currently focus on the use of antisense oligonucleotides (antagomirs) to sequester the miRNA or synthetic mimics to restore its function in deficient cells, though these remain largely in the preclinical or early clinical research phases (PubMed: 28813311).

Mechanism of action

Antisense inhibition of miRNA activity using antagomirs or locked nucleic acids (LNAs) to sequester the mature miRNA, and restoration of miRNA function using synthetic miRNA mimics to replace deficient levels (PubMed: 28813311).
