Overview

MicroRNA 2117 (miR-2117) is a small, non-coding RNA molecule involved in the regulation of gene expression at the post-transcriptional level. It is downregulated in liver cancer stem cells and is associated with poor prognosis in hepatocellular carcinoma patients. Functional studies show that miR-2117 suppresses self-renewal and tumorigenicity of liver cancer stem cells, primarily by targeting the stemness transcription factor SOX2. Overexpression of miR-2117 increases sensitivity of liver cancer cells to cisplatin and may serve as a biomarker for predicting response to transcatheter arterial chemoembolization (TACE) therapy. Due to its regulatory role in cancer stem cell expansion and drug resistance, miR-2117 is being investigated as a novel therapeutic target for hepatocellular carcinoma and as a prognostic biomarker.

Mechanism of action

Post-transcriptional gene silencing (via direct binding to the 3′-UTR of SOX2 mRNA, leading to its downregulation); Modulation of drug sensitivity (increases sensitivity to chemotherapy by modifying cancer stem cell populations).
