Overview

microRNA 219a-1 (MIR219A1) is a small, non-coding RNA gene located on chromosome 6, critical for oligodendrocyte differentiation and myelination in the central nervous system, and functions as a tumor suppressor in various cancers, including gliomas, breast, colorectal, ovarian, hepatocellular carcinoma, and gallbladder cancer[1][2]. MIR219A1, along with other precursors such as MIR219A2, encodes mature miR-219, which regulates gene expression post-transcriptionally by targeting mRNAs involved in cell proliferation, migration, apoptosis, and epigenetic networks. Dysregulation of MIR219A1 may contribute to neurodevelopmental disorders, tumor progression, and resistance to therapy, while also serving as a promising biomarker for disease prognosis and a therapeutic target[1][2].

Mechanism of action

Drugs/molecules can increase or decrease MIR219A1 expression, affecting cancer cell proliferation and apoptosis. Chemotherapy agents (e.g., cisplatin) may be more effective due to miR-219a-1–mediated sensitization. Epigenetic modulation (e.g., promoter methylation by triclosan) downregulating miR-219a-1 activity.
