In oncology, it exhibits a dual role, acting as a tumor suppressor in some cancers like hepatocellular carcinoma while functioning as an oncomiR in others such as certain types of breast cancer. Beyond malignancy, miR-22-3p is a key regulator in cardiovascular health, where its dysregulation is linked to cardiac hypertrophy and remodeling. Therapeutic strategies currently focus on using miRNA mimics to restore its function or antagomirs to inhibit its activity, though precise delivery and off-target effects remain significant hurdles in clinical development.

Mechanism of action

MicroRNA 22-3p functions primarily through the RNA-induced silencing complex (RISC) to bind to the 3' untranslated region (UTR) of target mRNAs, leading to translational repression or mRNA degradation. By downregulating specific oncogenes or tumor suppressors depending on the cellular context, it modulates various signaling pathways such as PTEN/AKT, Wnt/beta-catenin, and AMPK.
