Overview

MicroRNA-26a is a small non-coding RNA molecule that serves as a master post-transcriptional regulator of gene expression by binding to the 3' untranslated regions (UTRs) of multiple target mRNAs [1, 6]. It plays a dual role in human physiology, acting as a potent tumor suppressor in various malignancies—such as hepatocellular carcinoma, breast cancer, and melanoma—while also regulating critical processes in the cardiovascular and respiratory systems [4, 11, 12]. In many disease states, miR-26a is significantly downregulated, leading to the pathological overexpression of oncogenes and pro-fibrotic factors like EZH2, CTGF, and SMAD1 [5, 10, 16]. Therapeutic strategies currently under investigation include the use of miR-26a mimics to restore its function in cancer and heart failure, as well as miR-26a inhibitors (antagomirs) to modulate its activity in specific inflammatory or fibrotic contexts [2, 4, 27]. Additionally, miR-26a has been shown to enhance the sensitivity of cancer cells to conventional chemotherapeutic agents like docetaxel and cisplatin [11, 28]. Despite its therapeutic potential, clinical translation is hindered by challenges such as the need for efficient, tissue-specific delivery systems and the risk of off-target effects due to its broad regulatory network [2, 32, 35].

Mechanism of action

MicroRNA-26a functions by binding to the 3' untranslated regions (UTRs) of target messenger RNAs (mRNAs), leading to translational inhibition or mRNA degradation [6, 9]. It modulates multiple signaling pathways, including PI3K/AKT, TGF-beta/Smad, and Hedgehog, by targeting key regulators such as EZH2, PTEN, SMAD1, CTGF, and GSK3B [4, 5, 10, 11, 22].
