Overview

MicroRNA-27 is a highly conserved, small, non-coding RNA that post-transcriptionally regulates gene expression by binding to partially complementary sequences in the 3′ untranslated regions of target mRNAs, inducing translational repression or mRNA degradation. The miR-27 family consists of miR-27a and miR-27b, which are encoded by separate genomic loci as part of the Mir-23-27-24 clusters. miR-27 plays crucial roles in cellular proliferation, apoptosis, metabolism, adipogenesis, angiogenesis, and developmental processes. Dysregulated miR-27 expression is associated with a broad spectrum of disease states including cancer, fibrosis, cardiovascular, and metabolic diseases. Its activity in multiple essential pathways makes it a promising, but complex, therapeutic and biomarker candidate.

Mechanism of action

Antisense inhibition (anti-miR or LNA-mediated): Antisense oligonucleotides bind miR-27, preventing its interaction with target mRNAs. miRNA mimics: Synthetic miR-27 molecules supplement endogenous function, increasing post-transcriptional regulation of target genes. Translational repression: Modulators of miR-27 affect the translation (and sometimes subsequent stability) of specific target mRNAs such as HMGCR, EGFR, PHB, and PPARγ.
