Overview

miR-3065-5p-responsive messenger RNAs (mRNAs) are a collection of transcripts whose expression levels are controlled by the microRNA miR-3065-5p. This regulation occurs through the binding of the microRNA to complementary sequences, usually within the 3' untranslated region (UTR) of the target mRNAs (Source: PubMed ID 31480385). Once bound, the microRNA-induced silencing complex (miRISC) promotes mRNA degradation or prevents the translation of the mRNA into a functional protein. Validated responsive mRNAs for miR-3065-5p include those for CXCL12, SOX2, and YAP1, which are involved in critical pathways like cell growth and epithelial-mesenchymal transition (Source: PubMed ID 26109430, 30106431). In various malignancies, such as osteosarcoma and glioma, miR-3065-5p is frequently downregulated, leading to the overexpression of these responsive mRNAs and subsequent tumor progression. Therapeutic strategies aim to restore miR-3065-5p levels using synthetic mimics to downregulate these oncogenic mRNAs. However, the primary challenge in targeting these mRNAs collectively is the inherent lack of specificity, as a single microRNA can influence hundreds of different genes. This broad regulatory reach increases the risk of off-target effects in healthy tissues, complicating the development of safe and effective miRNA-based therapies.
