Overview

MicroRNA-30c-1 is a small non-coding RNA (~18–24 nucleotides) of the microRNA-30 family, encoded on human chromosome 1p34.2, that regulates gene expression post-transcriptionally by binding to complementary sequences in the 3' untranslated regions of target mRNAs, leading to mRNA degradation or translational repression. Through regulation of genes involved in cell cycle, apoptosis, EMT, and cytoskeletal dynamics, miR-30c-1 acts chiefly as a tumor suppressor in diverse cancers—by directly targeting genes such as Notch1, SOX9, SNAI1, VIM, TWF1, and others, it inhibits tumor cell proliferation, migration, invasion, and EMT, with its downregulation frequently associated with worse cancer prognosis. miR-30c-1 and its passenger strand (miR-30c-1-3p) are both biologically relevant, with evidence showing their anti-tumor effects and utility as prognostic biomarkers and potential therapeutic targets in human cancers.

Mechanism of action

As a therapeutic target, miR-30c-1 or mimics could suppress translation of oncogenes (such as Notch1, VIM, TWF1, SNAI1, SOX9, MTA1, KRAS, CCND2, and others) leading to inhibition of tumor growth, migration, invasion, and EMT. Antagomirs (inhibitors) can block its function and potentially reveal effects of endogenous miR-30c-1 deficiency.
