Overview

MicroRNA 3132 is an intronic microRNA transcribed as part of the human TMEM198 gene. It is a small non-coding RNA (about 18-22 nucleotides) whose main function is to regulate gene expression post-transcriptionally by binding to 3’ untranslated regions of mRNAs. Recent research shows that miR-3132 is downregulated in a wide range of cancer cell lines and that re-introduction (via synthetic miRNA mimics) potently suppresses cancer cell proliferation and induces apoptosis, particularly through upregulation of the pro-apoptotic ligand TRAIL and activation of Type I interferon/JAK-STAT signaling. This apoptosis is independent of p53 status and relies on extrinsic pathway markers. The therapeutic concept of miR-3132 is under pre-clinical investigation as a novel approach to cancer treatment.

miRNA mimics: Direct replacement of suppressed miR-3132, restoring its function in cancer cells. Induction of cell-surface TRAIL and activation of extrinsic apoptotic pathway. Upregulation of interferon and JAK-STAT signaling resulting in transcriptional activation of TRAIL and apoptosis.
