MicroRNA 3156-2 (MIR3156-2) is a small, non-protein-coding RNA gene that produces mature microRNAs, such as miR-3156-5p, which regulate the expression of various genes at the post-transcriptional level by binding to complementary sequences in mRNA transcripts. MIR3156-2 has been shown to be significantly downregulated in the serum of patients with multiple endocrine neoplasia type 1 (MEN1), a hereditary cancer syndrome, indicating a potential role as a circulating biomarker in this context. Its functional targets include genes like mortality factor 4-like 2 (MORF4L2), involved in transcriptional regulation and tumor biology, with experimental evidence for its regulatory effect in neuroendocrine tumor cells. There is currently no evidence to suggest that MIR3156-2 is a direct drug target or that any drugs act via modulation of its expression, but its role in disease (notably neuroendocrine tumors) and its use as a biomarker are areas of active study. MIR3156-2 encodes at least two mature miRNA products; miR-3156-5p is the best-characterized and is the subject of experimental and biomarker research. miRNAs, including MIR3156-2, function by guiding the RNA-induced silencing complex (RISC) to target mRNAs, mediating mRNA degradation or translational repression. Although described in cancer settings, there is no robust evidence that MIR3156-2 constitutes a direct molecular therapeutic target (e.g., receptor, enzyme, transporter), and it is more accurately classified as a genetic biomarker and a regulator rather than a druggable target.
