MicroRNA 3648-1 (MIR3648-1) is a human-specific non-coding microRNA involved in post-transcriptional gene regulation. It exerts its biological functions by binding to the untranslated regions of target mRNAs, resulting in mRNA degradation or translational repression. MIR3648-1 is implicated in the modulation of cell proliferation and tumor progression by targeting multiple tumor suppressors (including APC2 and SOCS2) and is associated with activation or inhibition of the Wnt/β-catenin signaling pathway, influencing cyclin D1/E1 and p21 levels. Its role varies by cancer type: it acts as an oncogene in lung, prostate, and bladder cancer, but may suppress tumor growth in gastric cancer. Elevated levels of MIR3648-1 are linked to higher proliferation, invasion, metastasis, and poor patient prognosis in several tumors.
miRNA inhibitors: synthetic molecules (e.g., antisense oligonucleotides) bind and inhibit MIR3648, thereby releasing downstream targets (such as SOCS2, APC2) from suppression and restoring tumor suppressor pathways. miRNA mimics: increase MIR3648 activity, used experimentally to decrease expression of target genes.
