Overview

MicroRNA 3661 (miR-3661) is a human non-coding RNA molecule belonging to the microRNA family, transcribed as a cis-antisense miRNA from the strand opposite to PPP2CA, the gene encoding the catalytic subunit alpha of protein phosphatase 2A (PP2A)[1]. It was first described in 2010 and shown to specifically and efficiently suppress the expression of PPP2CA at both mRNA and protein levels in human cells. As a result, miR-3661 increases the phosphorylation of PP2A substrates, such as AKT and GSK-3β, thus modulating critical signaling pathways involved in cell proliferation and survival[1]. Due to its regulation of a major cellular phosphatase with established cancer relevance, dysregulation of miR-3661 may have implications in cancer biology and potentially other disease contexts where PP2A function is perturbed[1]. There are currently no known drugs that directly target miR-3661, nor are there established uses as a clinical biomarker. Safety concerns would be similar to those of manipulating microRNAs or essential cellular phosphatases, with risk of disrupting key homeostatic processes.
