Overview

MicroRNA 374c (miR-374c) is a small, non-coding RNA molecule (microRNA) involved in the post-transcriptional regulation of gene expression via mRNA destabilization and inhibition of translation. It is part of the miR-374 family, a highly conserved group of miRNAs located in the X chromosome inactivation center as intronic sequences of the FTX long non-coding RNA. miR-374c and related family members act as pivotal regulators of cell growth, differentiation, apoptosis, and other key cell processes. Pathologically, altered miR-374 family expression contributes to several disease processes, most notably various cancers (e.g., liver, gastric, colorectal, pancreatic) where it influences tumor growth, metastasis, and therapeutic response. Because of its regulatory potency, miR-374c is of interest both as a biomarker and as a potential therapeutic target, though clinical translation is limited by specificity and delivery challenges.

Mechanism of action

Synthetic miRNA mimics can increase the activity of miR-374c, inducing downregulation of its gene targets and potentially inhibiting cancer cell proliferation or sensitizing cells to radiotherapy. Antisense inhibitors (antagomirs) or LNA-modified oligonucleotides can bind and inhibit miR-374c, blocking its regulatory effects, possibly reversing its function in oncogenesis or chemoresistance.
