Overview

MicroRNA 376c is a small, non-coding RNA molecule (miRNA) located in a syntenic cluster on human chromosome 14[2][3][4]. It functions as a post-transcriptional regulator by binding complementary sequences in target mRNAs, typically resulting in translational inhibition or mRNA destabilization via incorporation into the RNA-induced silencing complex (RISC)[4]. MIR376C has roles in regulating cell proliferation, apoptosis, cell cycle progression, and migration, largely through direct suppression of oncogenic/pro-survival or apoptotic pathway gene products such as CCND1, ALK7, ARID4A, RUNX2, and ARID2[2][3][5][7]. Its expression is dysregulated in numerous cancers—with either upregulation (promoting carcinogenesis and chemoresistance in some tumors such as gastric and ovarian cancer) or downregulation (tumor suppressor activity in others like neuroblastoma and melanoma)[2][3][5][7]. It also has utility as a diagnostic biomarker for cancer and hypoxic-ischemic injury[5][6]. Therapeutic targeting of miR-376c is still experimental, with potential but unaddressed safety and specificity risks.
