MicroRNA 377 (miR-377) is a short, non-coding RNA molecule that regulates the expression of target mRNAs at the post-transcriptional level, primarily by binding to complementary sites in the 3’ untranslated regions of target genes, leading to their translational repression or degradation[3]. miR-377 is implicated in multiple biological processes including cell proliferation, apoptosis, and angiogenesis. It functions predominantly as a tumor suppressor in several types of cancer, where its downregulation is often associated with increased malignancy and poor clinical outcome[1][4][6]. miR-377 directly targets oncogenic mRNAs such as astrocyte elevated gene-1 (AEG-1) in non-small-cell lung cancer[1], specificity protein 1 (Sp1) in glioblastoma[4], and TIAM1 in hepatocellular carcinoma[6], thereby inhibiting growth, invasion, and metastasis of cancer cells. Beyond oncology, miR-377 is involved in diabetic microvascular complications and modulates angiogenic and inflammatory pathways in the cardiovascular system[3][5]. It is considered a potential biomarker and therapeutic target, but clinical applications require caution due to its broad regulatory roles and potential for off-target effects.
