microRNA 378i (MIR378I) is a member of the microRNA-378 family, part of a class of small non-coding RNAs that regulate gene expression primarily by binding to target mRNA and inhibiting their translation or inducing degradation. The miR-378 family is implicated in the regulation of cell survival, proliferation, apoptosis, metabolic processes, and angiogenesis, with established roles in cancer and cardiovascular pathophysiology. While much functional and disease association data exist for other miR-378 variants (notably miR-378a), direct evidence for MIR378I’s unique roles is limited. Confusion in the miR-378 nomenclature has led to difficulties in interpreting and assigning specific biological and clinical relevance to miR-378i, and current literature does not clearly establish MIR378I as a singular therapeutic target distinct from its family members.
