Overview

microRNA 380 (MIR380) is a small, non-coding RNA (~20-24 nucleotides) of the microRNA family, encoded within the human genome as an ncRNA gene. It is involved in the post-transcriptional regulation of gene expression, usually by binding to complementary sequences in the 3′ untranslated region (UTR) of messenger RNAs, leading to their silencing either through translational inhibition or mRNA destabilization. In specific contexts, miR-380-3p suppresses translation of the Sp3 transcription factor, which contributes to cell proliferation inhibition and apoptosis induction in neural cells, and enhances neurotoxic effects induced by paraquat exposure. MIR380 expression is linked to various diseases, particularly cancers of the nervous system and potentially other neurological conditions. Therapeutically, while miRNAs like MIR380 are attractive molecular targets, there are currently no approved drugs targeting this miRNA; experimental approaches utilize oligonucleotide mimics or inhibitors to study or modulate its activity.

Mechanism of action

Synthetic miR-380-3p mimics repress translation of target mRNA, such as Sp3 transcription factor, leading to cell proliferation inhibition and increased apoptosis. Inhibitors block miR-380-3p activity, potentially permitting higher expression of targets like Sp3.
