MicroRNA 3928 (miR-3928) is a non-coding regulatory RNA belonging to the microRNA class, primarily expressed at lower levels in glioblastoma samples compared to healthy brain tissue[1]. It acts as a tumor suppressor miRNA, inhibiting cell growth, proliferation, and invasion, and inducing apoptosis in glioblastoma models. MiR-3928 directly targets mRNAs of multiple oncogenes, including MDM2 (negative p53 regulator), CD44, DDX3X, HMGA2, CCND1, BRAF, ATOH8, and BMI1, leading to their downregulation. This results in p53 activation and wide disruption of malignant signaling pathways, positioning miR-3928 as a master regulatory miRNA with therapeutic promise in cancer, particularly glioblastoma[1][2]. No drugs currently target miR-3928 directly, but its downregulation and regulatory profile lend itself as a biomarker and as a candidate for potential therapeutic intervention. The delivery, specificity, and broad target profile associated with miRNA molecules pose ongoing challenges for clinical development[1].
