Overview

MicroRNA 4327 (miR-4327) is a non-coding RNA molecule classified as a microRNA, which is involved in post-transcriptional regulation of gene expression by binding to complementary sequences in the 3' untranslated region (UTR) of target messenger RNAs. In human osteoblasts, miR-4327 has been shown to directly target the CREB-binding protein (CBP), thereby modulating the acetylation and stability of Runx2, a critical transcription factor for bone formation and remodeling. TGF-β1 stimulation leads to a decrease in miR-4327 expression, resulting in elevated CBP and MMP-13 (matrix metalloproteinase-13) levels. Overexpression of miR-4327 reverses this effect, reducing MMP-13 levels via CBP-mediated Runx2 acetylation. This regulatory axis is essential for bone homeostasis, and its disruption is associated with abnormal bone remodeling, which may contribute to diseases such as osteoporosis and osteoarthritis. Currently, miR-4327 is not recognized as a direct therapeutic target, receptor, or enzyme; rather, it acts as a regulatory RNA molecule influencing key protein interactions in bone biology[1][2].
