MicroRNA 433 (MIR433) is a short non-coding RNA molecule involved in the post-transcriptional regulation of gene expression through mRNA binding and silencing. It is transcribed as part of the imprinted microRNA cluster at chromosomal region 14q32.2 and matures into active forms hsa-miR-433-3p and hsa-miR-433-5p. MIR433 functions predominantly as a tumor suppressor in diverse cancers by inhibiting proliferation, migration, invasion, cell cycle progression, and promoting apoptosis through targeting oncogenic genes such as c-Met, CREB1, HDAC6, and Notch1. Its expression is often silenced epigenetically in cancers via promoter hypermethylation, and restoration of MIR433 levels suppresses tumor progression and EMT processes. Beyond oncology, MIR433 is implicated in neurodegenerative and inflammatory diseases, and is considered both a therapeutic target and a potential biomarker for prognosis and treatment selection.
