microRNA 4458 (miR-4458) is a small, non-coding RNA molecule belonging to the microRNA family that regulates post-transcriptional gene expression by binding to target mRNA sequences, typically reducing their expression. It acts as a tumor suppressor in several cancer types (notably lung, colon, and hepatocellular carcinoma) by inhibiting key oncogenes including IGF1R, CCND1, and Lin28B. Increased expression of miR-4458 represses cancer cell proliferation, induces cell cycle arrest at G0/G1 phase, and promotes apoptosis, while reduced levels are associated with poor clinical outcomes in lung cancer. Its modulation has potential as both a therapeutic strategy and diagnostic/prognostic biomarker in oncology, although there are currently no approved drugs directly targeting this miRNA, and therapeutic targeting is challenged by specificity and delivery issues common to miRNA-based interventions.
