Overview

MicroRNA 4474 (miR-4474, hsa-mir-4474) is a small, non-coding RNA molecule that functions as a post-transcriptional regulator of gene expression by binding to complementary sequences on target mRNAs, leading to translational repression or degradation. MiR-4474 has been shown to be upregulated in colorectal cancer (CRC) tissues infected with *Fusobacterium nucleatum*, and directly targets CREB-binding protein (CREBBP) mRNA, resulting in reduced CREBBP expression. This modulation of CREBBP impacts Wnt/β-catenin signaling and cell proliferation, which may contribute to colorectal carcinogenesis and tumor progression. No drugs are known to directly interact with miR-4474, and its clinical utility as a biomarker or therapeutic target remains experimental.

Biological functions

Post-transcriptional regulation of gene expressionNegative regulation of target mRNA (notably CREBBP mRNA)Regulation of cell proliferationInvolvement in colorectal cancer progression, particularly in response to Fusobacterium nucleatum infection
03

Disease associations

Cancer (specifically colorectal cancer (CRC) associated with *Fusobacterium nucleatum* infection)Other: Potentially in oncogenic pathways by modulating tumor suppressor/coactivator expression
04

Safety considerations

Specific safety concerns related to targeting miR-4474 are unknown and not currently established in the literature.Generally, modulating miRNAs systemically can have off-target genetic regulatory effects due to their involvement in complex networks
05

Biomarkers

miR-4474 expression in tissue may serve as a biomarker for *F. nucleatum*-positive colorectal cancer and malignant progression, but this is not yet clinically validatedCREBBP expression changes as a downstream readout of miR-4474 modulation
