MicroRNA 4512 is a small non-coding RNA molecule of the microRNA class, primarily identified as a human regulatory RNA (hsa-miR-4512). It functions in the post-transcriptional regulation of gene expression, with a specific role in modulating innate immune responses. In systemic lupus erythematosus (SLE), miR-4512 is downregulated in monocytes and macrophages, which leads to increased expression of pro-inflammatory mediators such as TLR4 and CXCL2, thereby promoting cytokine secretion and neutrophil extracellular trap (NET) formation—a process implicated in SLE pathogenesis. miR-4512 acts by directly targeting the mRNAs of TLR4 and CXCL2, suppressing their translation. The abnormal regulation of this microRNA is associated with autoimmune and inflammatory mechanisms, primarily within the context of SLE pathophysiology. While no currently approved drugs interact directly with miR-4512, it is considered a novel therapeutic target and a potential biomarker for autoimmune activity and treatment response monitoring in SLE[2].
