Overview

microRNA 4534 (miR-4534) is a small (20–24 nucleotides) non-coding RNA molecule that regulates gene expression by binding to complementary sequences in target mRNAs, leading to translational inhibition or degradation of the target mRNA[6][10]. It has been found to act as an oncogenic microRNA in prostate cancer by directly targeting and downregulating PTEN, a tumor suppressor gene involved in the PI3K/Akt signaling pathway, contributing to increased cell proliferation, migration, and survival[2][7]. miR-4534 is upregulated in prostate cancer cells versus normal tissues and is associated with poor clinical outcomes[2][7]. It has also been identified as a urinary exosomal biomarker in diabetic kidney disease, where it may influence the FoxO signaling pathway by targeting BNIP3[1]. Therapeutic or biomarker approaches involving miR-4534 face challenges typical of microRNA-based strategies, including specificity and off-target effects.

Mechanism of action

Downregulation of tumor suppressor genes (e.g., PTEN) via direct binding to messenger RNA, reducing its translation or triggering its degradation[2][7]

03

Biological functions

Regulation of gene expression (post-transcriptional)Cell cycle regulationCell proliferationApoptosis
04

Disease associations

Cancer (specifically, prostate cancer)Diabetic kidney disease
05

Safety considerations

Potential for off-target effects if therapeutically modulated, as microRNAs can target multiple mRNAsChallenges in biomarker specificity due to the broad regulatory roles of microRNAs across different tissues and diseases
06

Biomarkers

Diagnostic biomarker for prostate cancer (due to upregulation and association with poor prognosis)[2][7]Urinary biomarker for progression of diabetic kidney disease[1]
