Overview

MicroRNA 4649 is a small non-coding RNA molecule that negatively regulates gene expression post-transcriptionally, primarily by binding to specific mRNA targets and repressing their translation. In TNBC, miR-4649-5p is expressed at low endogenous levels, but higher expression is associated with improved patient survival. Experimental overexpression of miR-4649-5p reduces tumor cell growth, proliferation, and migration. Its tumor-suppressive activity is largely mediated through direct targeting and downregulation of phosphatidylinositol-4-phosphate 5-kinase type 1 gamma (PIP5K1C), thereby inhibiting the PI3K/AKT signaling axis—a key pathway promoting cancer cell survival and migration. Therapies that enhance miR-4649-5p function, or combine its upregulation with inhibitors of PIP5K1C or AKT, may have synergistic anti-cancer effects. miR-4649-5p shows promise as both a therapeutic target and a prognostic biomarker in oncology.
