microRNA 4677 (miR-4677)

Target
miR-4677
Molecular classification
microRNA, Non-coding RNA (ncRNA), Regulatory RNA
01

Overview

microRNA 4677 is a small, non-coding RNA molecule classified as a microRNA, transcribed from the human genome and processed by the microRNA biogenesis pathway. It regulates gene expression by incomplete complementary binding to the 3' untranslated regions of target mRNAs, resulting in suppression of translation or degradation of these mRNAs. miR-4677-3p, a specific mature form, is frequently downregulated in gastric cancer and other tumors, where its overexpression suppresses proliferation, migration, and invasion of cancer cells, primarily through inhibition of CEMIP and modulation of PI3K/AKT signaling. miR-4677’s levels are being studied as biomarkers for cancer diagnosis and prognosis, and it is proposed as a potential therapeutic target in oncology[1][3][4].

Mechanism of action

Drugs such as PI3K/AKT inhibitors block proliferation and metastasis downstream of the miR-4677 axis by counteracting the pathway promoted by reduced miR-4677 expression. miRNA mimics or inhibitors modulate cellular levels of miR-4677, thereby altering cancer cell behavior in preclinical models.

03

Biological functions

Regulation of gene expression at the post-transcriptional level by binding to the 3' untranslated regions (3' UTR) of mRNAs, typically resulting in mRNA degradation or inhibition of translationInhibition of cell proliferation, migration, and invasion in cancer cells, notably gastric cancer and lung adenocarcinoma, via targeting specific mRNAs such as CEMIP and ZEB1Modulation of signaling pathways including PI3K/AKT
04

Disease associations

Cancer (gastric cancer, lung adenocarcinoma, peripheral T-cell lymphoma, oral squamous cell carcinoma)Other (potentially relevant in additional cancers based on microRNA family associations)
05

Safety considerations

Challenges include delivery, off-target effects, and specificity in using miRNA mimics or inhibitors therapeutically.Specific safety concerns for miR-4677 not detailed in search results.
06

Biomarkers

Downregulation of miR-4677-3p in tumor tissues (gastric and other cancers) may serve as a diagnostic or prognostic biomarkerCorrelation of miR-4677-3p and CEMIP expression in tumor tissues
