MicroRNA 4680 (MIR4680)

Target
MIR4680
Molecular classification
microRNA (miRNA), Non-coding RNA
01

Overview

MicroRNA 4680 (MIR4680) is a short, non-coding RNA (miRNA) whose mature form mediates post-transcriptional gene silencing by binding target mRNAs and inhibiting their translation or promoting degradation. In human embryonic palatal mesenchymal (HEPM) cells, MIR4680 is upregulated by environmental agents such as *all-trans retinoic acid* and *phenytoin*, leading to suppressed proliferation and downregulation of key genes required for normal palate development, including *ERBB2* and *JADE1*. These findings have implicated MIR4680 as a potential genetic and pharmacological target in the study and prevention of congenital cleft palate

02

Mechanism of action

Drug-induced upregulation of MIR4680 suppresses cell proliferation by downregulating ERBB2 (a receptor tyrosine kinase), JADE1 (a nuclear protein), and other predicted targets

03

Biological functions

Post-transcriptional regulation of gene expressionNegative regulation of cell proliferationRegulation of genes involved in palate morphogenesis
04

Disease associations

Cleft palate / craniofacial developmental abnormalitiesPotentially relevant in birth defects caused by environmental or drug-induced factors (e.g., retinoic acid, phenytoin)
05

Safety considerations

Overexpression or activation may cause abnormal palate development (cleft palate)Altered expression has implications for developmental toxicity in response to drugs or environmental factors
06

Interacting drugs

All-trans retinoic acid (atRA)

07

Biomarkers

MIR4680 expression can be used to monitor risk for cleft palate or response to teratogenic compounds in model systems
