Overview

MicroRNA 4728 (miR-4728, also known as MIR4728 or hsa-miR-4728) is a non-coding RNA molecule located within an intron of the ERBB2/HER2 gene. It acts as a tumor-suppressive microRNA by negatively regulating the MAPK signaling pathway, particularly through direct inhibition of MST4 kinase, thereby suppressing ERK pathway activation. miR-4728 has been shown to inhibit cancer cell proliferation, migration, and invasion, and its expression is commonly downregulated in several cancer types, including breast cancer, non-small cell lung cancer, Burkitt lymphoma, and papillary thyroid cancer[1][2][3][4][5]. miR-4728 may also be used as a diagnostic and prognostic biomarker, correlating with tumor stage, size, metastasis, and HER2 status, and represents a novel example of an intronic miRNA that can suppress the oncogenic effects of its host gene[3][4]. There are currently no approved drugs directly targeting miR-4728, but it remains a promising candidate for RNA-based therapeutic strategies.
