Overview

MicroRNA 487b (miR-487b) is a small (~22 nucleotide) non-coding RNA belonging to the miRNA class, transcribed from the MIR487B gene. It functions post-transcriptionally by binding target mRNAs, suppressing their translation and/or triggering degradation. miR-487b directly regulates multiple oncogenes and cancer pathway genes (KRAS, MYC, SUZ12, BMI1, WNT5A), antagonizing cell proliferation and invasion. It acts as a tumor suppressor, with decreased expression linked to cancer metastasis and poor prognosis in colorectal cancer, lung cancer, and neuroblastoma. It is under clinical investigation as both a therapeutic (using mimics) and diagnostic (biomarker) target. Its broad regulatory role means mimics/targeting agents may have complex multi-pathway effects, presenting both therapeutic promise and safety concerns[1][2][3][5].

Mechanism of action

miR-487b mimics: Restore/supplement tumor-suppressor activity, leading to suppression of target oncogene expression (such as KRAS, MYC, SUZ12, BMI1, WNT5A), downregulation of involved pathways (MAPK/ERK, AKT, WNT/β-catenin), and inhibition of proliferation and metastasis. miR-487b inhibitors: Potentially promote cell growth and invasion, opposite to its usual action.
