Overview

MicroRNA 497 (miR-497) is a small non-coding RNA molecule (~22 nucleotides) encoded by the first intron of the MIR497HG gene on chromosome 17p13.1. As a member of the highly conserved miR-15/16/195/424/497 family, miR-497 functions in post-transcriptional gene silencing by binding to the 3′-untranslated region (3′UTR) of target mRNAs through its seed sequence (AGCAGCA), thereby reducing mRNA stability or inhibiting translation. miR-497 is expressed in normal tissues but is frequently dysregulated in multiple cancers, most commonly downregulated and acting as a tumor suppressor, though it can act as an oncogene in particular contexts such as gliomas. Its biological functions include inhibition of cell proliferation, induction of apoptosis, cell cycle arrest, anti-angiogenesis, and modulation of invasion and metastasis. Therapeutically, miR-497 is being explored as a target for cancer treatment and as a non-invasive biomarker for patient selection and efficacy monitoring. However, its pleiotropic roles and complex gene network interactions present safety and specificity challenges for clinical application.
