MicroRNA 509-2 (miR-509-2) is a regulatory non-coding RNA molecule classified as a microRNA, first identified in Homo sapiens. It acts as a tumor suppressor in multiple cancer types by post-transcriptional repression of target mRNAs. Its downregulation has been reported in several cancers, including tongue squamous cell carcinoma (TSCC), renal carcinoma, ovarian cancer, gastric cancer, and triple-negative breast cancer. In TSCC specifically, miR-509-2 inhibits cell proliferation and invasion by directly targeting the epidermal growth factor receptor (EGFR) and suppressing EGFR-associated signaling pathways, including phosphorylated AKT and ERK. Restoration of miR-509-2 expression can suppress tumor cell growth, migration, and invasion, and improve sensitivity to chemotherapeutic agents like cisplatin in resistant cancers. The miR-509-2/EGFR regulatory axis constitutes a promising area for potential cancer therapeutics and biomarker development[1].
