Overview

MicroRNA-511-5p (miR-511-5p) is a highly conserved small non-coding RNA that functions as a critical post-transcriptional regulator of gene expression (PMID: 22504410). It is uniquely encoded within the fifth intron of the MRC1 gene, which encodes the mannose receptor (CD206), and is predominantly expressed in M2-polarized macrophages where it modulates the inflammatory phenotype (PMID: 24613947). miR-511-5p exerts its biological effects by binding to the 3' untranslated regions (UTRs) of specific target mRNAs, such as Toll-like receptor 4 (TLR4) and Tribbles pseudokinase 2 (TRIB2), leading to their degradation or translational inhibition (PMID: 24613947, PMID: 28656231). In oncology, miR-511-5p typically acts as a tumor suppressor, with downregulated levels observed in colorectal cancer, glioblastoma, and hepatocellular carcinoma, where its restoration inhibits cell proliferation and migration (PMID: 26918253, PMID: 31233143). Conversely, its role in asthma involves the negative regulation of TLR4, suggesting that its expression levels can influence airway hyperresponsiveness and inflammation (PMID: 24613947). Therapeutic development targeting miR-511-5p involves the use of synthetic miRNA mimics to enhance its tumor-suppressive activity or antagomirs to modulate immune responses in inflammatory diseases. As a potential biomarker, its expression profile in tissue and biofluids is being investigated for diagnostic and prognostic utility in various malignancies and chronic inflammatory conditions.
