Overview

MicroRNA-524 (miR-524, hsa-miR-524) is a short non-coding RNA molecule (microRNA) that regulates gene expression post-transcriptionally by binding to target mRNAs and promoting their degradation or inhibiting their translation. It is transcribed as part of a longer RNA precursor and then processed to its mature, functional form. MiR-524-5p plays a role in suppressing tumor cell proliferation, migration, and invasion in multiple cancers by targeting key components of signaling pathways such as MAPK/ERK (notably BRAF and ERK2) and Notch (via NUMB). It acts as a tumor suppressor in glioma and melanoma, and its downregulation is associated with poorer outcomes and higher tumor grade in glioma. The molecule shows context-dependent functions: it is downregulated in some cancers (glioma, thyroid cancer) where it suppresses tumor progression, but upregulated in others (osteosarcoma) where it can promote cell proliferation. MiR-524-5p is being explored as both a therapeutic candidate and a prognostic biomarker, particularly in brain tumors.
