MicroRNA 548a-3 (miR-548a-3p) is a member of the mir-548 family of small, non-coding RNAs (microRNAs) that regulate gene expression by inhibiting translation or destabilizing target mRNAs. Like other microRNAs, it is produced as a 70 nucleotide precursor (pre-miRNA) from longer transcripts before being processed into its mature, active ~22 nucleotide form. miR-548a-3p has been shown to regulate diverse cellular pathways, notably those involved in immune cell function, cell proliferation, and apoptotic processes through its targeting of genes such as AKT2, PIK3R1, PIK3R3, and PIK3CB. It has clinical relevance as a candidate biomarker: for instance, elevated levels of serum miR-548a-3p are associated with favorable responses (no evidence of disease activity, NEDA-3) to fingolimod in patients with relapsing-remitting multiple sclerosis[1]. Dysregulation of miR-548a-3p has also been reported in autoimmune diseases including rheumatoid arthritis and psoriasis, as well as in acute graft-versus-host disease[1]. This molecule itself is not a classical therapeutic target like a receptor or enzyme, but rather acts as a regulatory RNA involved in disease pathogenesis and as a biomarker predictive of drug response[1][2]. Context on classification: MicroRNAs generally are not direct drug targets but serve important roles as biomarkers or indirect regulators; therefore, "is_target" is set to false and "is_incorrect" is true under definitions where a "target" refers to classical druggable protein entities (receptor, enzyme, transporter, etc.)[2].
