MicroRNA 548ac (miR-548ac) is a member of the microRNA family, encoded by the MIR548AC gene located within the first intron of the CD58 gene in humans. As a microRNA, miR-548ac is a non-coding RNA molecule approximately 20-24 nucleotides in length, functioning to regulate gene expression post-transcriptionally by binding to target mRNAs and promoting their degradation or inhibiting their translation. miR-548ac has been implicated in various cellular processes including inflammatory response regulation and cellular stress adaptation. Genetic variants affecting miR-548ac levels, particularly SNPs in its stem-loop region, are associated with altered risk of multiple sclerosis by modifying the expression of both miR-548ac and its host gene CD58 in immune cells. Experimentally, miR-548ac plays roles in modulating the transition of endothelial cells to mesenchymal states, notably under acidic stress by targeting proteins such as YB-1 and Snail, pathways relevant to cancer progression and metastasis. Its dysregulation is seen in certain cancers and neuroinflammatory disorders, making it a potential disease biomarker but not a direct drug target.
