Overview

MicroRNA 548b is a small non-coding RNA molecule (20–24 nucleotides) categorized within the miR-548 family. It regulates gene expression at the post-transcriptional level by binding to complementary sequences in target mRNAs, resulting in their degradation or translational suppression. miR-548b is frequently downregulated in several malignancies, notably hepatocellular carcinoma, where its loss is associated with aggressive tumor features. Restoration of miR-548b inhibits tumor cell proliferation, colony formation, migration, invasion, and activates apoptosis. Mechanistically, miR-548b targets high-mobility group box 1 (HMGB1) and suppresses PI3K–AKT pathway activation. It is proposed as a candidate biomarker and therapeutic target for cancer diagnosis and treatment, especially in liver cancer and primary CNS lymphoma. There are no major issues with the target’s designation. It is a well-characterized microRNA, but not a receptor, enzyme, channel, or transporter; its therapeutic target status relates to RNA-based interventions rather than classical pharmacology.

Mechanism of action

Therapeutic strategies may include miRNA mimics or inhibitors (antagomirs) to restore or block its function. Downregulates specific mRNA targets by binding to their 3′-UTR, causing transcript degradation or translation inhibition (e.g., HMGB1, MTA2).
