MicroRNA 548c (MIR548C) is a primate-specific member of the microRNA gene family involved in post-transcriptional gene regulation[3]. It is transcribed by RNA polymerase II, processed from primary hairpin transcripts by Drosha and Dicer, then incorporated into the RNA-induced silencing complex (RISC), which targets mRNAs for translational repression or destabilization[3]. MIR548C has two main mature forms, miR-548c-3p and miR-548c-5p, each functioning in different cellular contexts. Experimental studies show miR-548c-3p is downregulated in various cancers (including breast cancer and glioma), where it suppresses cell proliferation, migration, invasion, and induces apoptosis by targeting genes such as E2F3 and c-Myb[1][2]. miR-548c-5p can additionally regulate gene transcriptional activation in the nucleus, for example by increasing FOXC2 expression[4]. MIR548C is under investigation as a tumor suppressor and potential biomarker, but is not currently targeted by approved drugs. Caution is warranted in drug development owing to its broad involvement in gene regulatory systems and primate-specific nature[1][2][3][4].
