Overview

microRNA 548l (miR-548l) is a member of the non-coding microRNA family endogenously expressed in humans, encoded by the MIR548L gene. It acts as a regulatory RNA molecule, playing a significant role in cancer biology, especially in non-small cell lung cancer (NSCLC). miR-548l regulates important cellular functions, particularly by targeting the AKT1 gene involved in the PI3K/AKT/mTOR signaling pathway, which affects cancer cell proliferation, migration, invasion, and metastasis. Overexpression of miR-548l suppresses NSCLC cell migration and invasion, while a low level of miR-548l is associated with lymph node metastasis. Its regulatory effects are mediated, in part, by downregulation of AKT1, which leads to decreased mTOR, MMP-2, and MMP-9 expression, all important for cancer aggression and spread. Current data suggests miR-548l is not a druggable therapeutic target itself (e.g., not a receptor, enzyme, transporter), but rather a regulatory RNA with potential as a biomarker or indirect therapeutic node[1].
