MicroRNA-548x is a member of the primate-specific miR-548 family of non-coding RNAs, functioning primarily as a regulatory microRNA involved in post-transcriptional control of gene expression[2][3]. miR-548x directly targets multiple components of the PI3K/AKT/mTOR pathway, thereby suppressing growth and proliferation of human glioblastoma cell lines when overexpressed[2]. Downregulation or loss of miR-548x activity is associated with increased cell proliferation and may contribute to tumorigenesis. The miR-548 family is large and generally considered poorly conserved, but members have been implicated in both tumor suppressor and promoter activities in different cancers, with miR-548x showing evidence of tumor-suppressive effects specifically through modulation of growth-regulatory signaling cascades[2][3]. The molecule is under investigation as a potential biomarker and therapeutic agent, particularly for glioblastoma and possibly other cancers.
miR-548x acts as a tumor suppressor by directly downregulating several cancer-associated mRNAs involved in the PI3K/AKT/mTOR signaling pathway, including: - PI3KCB - AKT1 - mTOR - Rheb - CREB1 This downregulation results in impaired cell proliferation and cell cycle progression in cancer cells[2].
