Overview

MicroRNA 5581 (miR-5581) is a small, non-coding RNA molecule classified as a microRNA, involved in post-transcriptional regulation of gene expression through base-pairing with target messenger RNAs (mRNAs), leading to mRNA degradation or translational inhibition. miR-5581 has been identified with two major mature strands: miR-5581-3p and miR-5581-5p. Both have been described as tumor suppressors in several cancer models, notably in bladder cancer and mesothelioma, where overexpression inhibits cellular proliferation, migration, and epithelial-mesenchymal transition. miR-5581-3p exerts its effects by directly targeting oncogenes and critical pathways, such as SMAD3 (regulator of EMT) and FTO (an RNA demethylase). Its dysregulation is associated with disease states, particularly cancer, where its reduced expression correlates with increased tumor aggressiveness and poor prognosis. Currently, no drugs are clinically approved to modulate miR-5581, but its function as a gene expression regulator and emerging evidence as a biomarker make it a potential therapeutic target in oncology.

Mechanism of action

No conventional small molecule or biologic drugs known to directly target miR-5581; however, strategies such as miRNA mimics or inhibitors could modulate its function (general to microRNA therapeutics). Overexpression of miR-5581-3p downregulates SMAD3 and FTO, key molecules in tumor progression and RNA demethylation respectively. Sponging/antagonizing by long non-coding RNAs (e.g., LINC00526).
